The Blue-eyed Samurai is unlocked when you complete Cherry Blossom Viewing in Daigo.

This is the shortest main mission in the game. When the mission starts you are immediately in the boss room. You are facing William, the main protagonist who you played in the first Nioh game. All you have to do to finish the mission is defeating William. The fight will be won when his health reaches 30% (meaning there’s no need to deplete all of his health).
William Boss Fight
This fight is quite easy. Let William run towards you. While he’s on the move you do a single heavy attack in high-stance. He can’t defend while he’s running. If he does anything else you run sideways. William doesn’t have a lot of health so this fight won’t take long.
After defeating William, go talk to the girl Mumyo and the mission will complete.
This finishes The Blue-eyed Samurai main mission in Nioh 2. Now the mission In the Eye of the Beholder becomes available.
