The Sister Betrayed is unlocked when you’ve completed The Conspirator and Wolves at the Gates

Rewards: Minor Legend Increase, Adachi Half Mask, Minor Stealth Charm
Mission Info: (Masako Tale 9 of 9). Lady Mask has tracked her sister to clan Kikuchi’s old fort. We will make our way there and confront her. mask appears determine to do what she must. But her sister is the only family she has left… a sacrifice that will bring honour, but with it a great burden.
Objectives:
- Find Mask at Fort Kaminodake
- Follow Masako
- Follow Masako to the Kikuchi Estate
- Open the Gate
- Defeat the Samuari
- Follow Masako
- Defeat the Samuari
- Search for Masako
Starting Location: Fort Kaminodake
Find Masako at Fort Kaminodake
Ride to Fort Kaminodake to speak with Lady Tamako. Lady Tamako can be found preying at an altar under the wooden bridge of the Fort.
Follow Masako
Follow Masako to your horse and ride with her to the Kikuchi estate.
Follow Masako to the Kikuchi Estate
When prompted, dismount your horse and follow Masako to the Kikuchi estate on foot.
Open the Gate
Now get rid of the few enemies in front of the gate to the estate and then open it.
Defeat the Samurai
After the next brief cutscene, you’ll have to get rid of the various samurai surrounding the estate.
Follow Masako
Next follow Masako up the hill where you’ll face Lady Hana, Masako’s sister.
Defeat the Samurai
Here you’ll be welcomed by a second wave of samurai. Defeat them all to proceed.
Search for Masako
Lady Masako is confronting her sister. Use Focused Hearing to locate and reach her. The following cutscene will conclude Lady Masako’s tale.
This finishes the The Sister Betrayed quest in Ghost of Tsushima and the whole Lady Masako’s tale. Completing Lady Masako’s tale will unlock 
The Vengeful Warrior.
